This is a special SAXS/DEER target. The aim is to compare submitted multistate models against SAXS and DEER data generated by Rick Page (Miami University) and Susan Tsutakawa (Lawrence Berkeley National Laboratory). The ubiquitin ligase CHIP is critical to protein quality control, as it recognizes and ubiquitinates Hsp70-chaperoned client proteins to target them for degradation [Qian et al. CHIP-mediated stress recovery by sequential ubiquitination of substrates and Hsp70. Nature. 2006 440:551-5. doi: 10.1038/nature04600]. The structural dynamics of the CHIP/Hsp70 complex are essential for this function, as current models suggest that conformational ensembles are required to facilitate the interaction with the E2-ubiquitin conjugate and ensure specificity in lysine targeting.
Predictors are provided the amino acid sequences for a two-polypeptide complex system consisting of the CHIP-TPR domain and the Hsp70 SBD+Lid+Tail domains, which interact with a 1:1 stoichiometry. PDB structures for individual domains are available as starting templates (e.g., PDB IDs 2c2l, 9o7d, 9o7e, 9o7f, 4b9a, 4b9q, 2kho, 7kru).
For the CASP submission, participants must provide an ensemble of 1 to 50 all-atom models that include all residues specified in the sequence; incomplete models will not be evaluated. The coordinate models should be supplemented by a populations.txt file listing each model of the ensemble with the corresponding relative population, expressed as a positive rational number. The sum of all populations should equal 1.0. All conformational ensembles are to be submitted through the dedicated CASP17 gateway https://predictioncenter.org/casp17/predictions_submission_ENSMBL.cgi, which provides further instructions on preparing submissions.
We ask participants to summarize the methods used for their submission (e.g., AI model, MD). The submitted model(s) will be assessed against SAXS and DEER data. The domains will not be evaluated at the atomic level. |
