Time-resolved crystallography has been used to obtain a series of structures spanning the catalytic cycle of a class A beta-lactamase, CTX-M-14. This target requires modeling of the covalent acyl-intermediate in the cleavage of dicloxacillin. Models will be compared with the corresponding experimental complex. There are structures of trapped covalent intermediates for beta-lactamases in the PDB, obtained by introducing mutations in the enzyme. What is different here is the time sampling, and the specific substrate. We expect time-resolved data to be more common in future, and include this challenge on a trial basis to encourage the implementation of appropriate methods, to determine what sort of issues may arise, and to promote discussion. |
