The MARTX effector domain Ras_Rap1-specific endopeptidase (RRSP) has been directly linked to highly virulent isolates of Vibrio vulnificus, a motile Gram-negative opportunistic human pathogen that causes rapidly progressing, life-threatening sepsis from ingesting contaminated seafood or exposing open wounds to seawater. V. vulnificus strains producing a MARTXVv toxin that carries RRSP show about 50-fold increased virulence in mice. This potent cytotoxicity of RRSP is due to its direct inhibition of the mitogen-activated protein kinases pathway (MAPK) by proteolytic processing of the small guanosine triphosphatases (GTPases) Ras and Rap1. Processing of Ras prevents the phosphorylation and activation of extracellular signal_regulated kinases (ERK). RRSP specifically recognizes the Switch I of Ras and Rap1 and then cleaves the Switch I between the Y32 and D33 residues. This activity is also found in toxins of other bacteria, including Photorhabdus spp. insect pathogens and Aeromonas spp. pathogens of fish.
