DpdA is annotated in the genomic databases as a tRNA guanine transglycosylase. However, its true function was determined to be the insertion of preQ0, a stable precursor to the modified nucleoside queuosine in DNA of bacteria and phage. Queuosine is a 7-deazaguanosine modification of the wobble position of tRNAs and has been studied extensively by us and other groups. Its recent discovery in the DNA of bacteria and phage is novel, and opens the field of DNA modification to studies beyond the known methylation of DNA, and highlights the cross-talk between RNA and DNA metabolism. Preliminary results show that queuosine modification of DNA by DpdA confers resistance against foreign endonucleoases, implying an ancient role in restriction-modification and defense against foreign DNA.
